Improving sperm DNA to improve fertilization rates in assisted reproduction

The success of Assisted Reproductive Techniques (ART) is heavily dependent on sperm DNA quality, which can be damaged by preparation methods and underlying reversible male conditions like varicocele and infection.

Assistant reproduction is defined as using advanced fertility techniques such as intrauterine insemination or in vitro fertilization to enhance fertilization rates. The success of these techniques relies on the ability to place the sperm or the sperm DNA in proximity to the unfertilized egg.

These techniques have provided fertility success for couples with significantly abnormal semen analysis with low count, low motility or very few sperm available through sperm extraction.

With the newfound success of these techniques also comes the realization that many couples are still not able to conceive after assisted reproduction.

Sperm DNA and sperm DNA quality have been suspected as leading to fertilization failure with assisted reproduction. Several new studies have looked at the sperm preparation technique to determine if better quality sperm can be made available for fertilization.

Density centrifugation, swim up separation, and microfluids sorting are all sperm preparation techniques being investigated to increase fertilization rates.

Excessive agitation of the sperm during preparation has been found to create free oxygen radicals and sperm DNA damage. This has limited the success of some sperm preparation techniques.
